Zebrafish (Danio rerio) ecotoxicological ABCB4, ABCC1 and ABCG2a gene promoters depict spatiotemporal xenobiotic multidrug resistance properties against environmental pollutants

چکیده

Marine organisms are naturally equipped with multixenobiotic resistance mechanisms that often governed by ATP-binding cassette (ABC) transporter family members. Previous studies focused on the target genes of ABC but little is known about functionality their promoter regions. Due to importance promoters in gene regulation, we functionally characterized three major xenobiotic zebrafish, namely ABCB4, ABCC1 and ABCG2a via silico transcription factor binding analysis vivo spatiotemporal expression analysis. The former revealed functional contributors (such as AP-1, C/EBP beta, HNF-1 NF-1 TFBSs) towards activity enhancement across four different tissues (liver, muscle, cell cycle immune cells) where majority them discovered were liver-specific whereas latter unearthed localization these at liver intestinal tracts during late embryogenesis (48, 72 96 hpf). This study contributes future ecotoxicology zebrafish.

Regulation of hepatic abcb4 and cyp3a65 gene expression and multidrug/multixenobiotic resistance (MDR/MXR) functional activity in the model teleost, Danio rerio (zebrafish).

Multidrug/multixenobiotic resistance (MDR/MXR) confers resistance to a diverse range of potentially toxic pharmaceuticals and environmental contaminants through a cellular response that involves the coordinated induction and activity of the ATP-binding cassette (ABC) transporter P-glycoprotein (P-gp) and the Phase I metabolizing enzyme cytochrome P450 3A (CYP3A). In mammals, ligand-mediated pre...

Danio Rerio, the Zebrafish

The zebrafish has long been a favourite model for the study of vertebrate development. Here we provide an overview of the current state of knowledge and resources for the study of this fish, with comments on the future direction of zebrafish genomics from Professor Mark Fishman and Dr Stephen Wilson.

BIOLOGY OF ZEBRAFISH, Danio rerio

Zebrafish, Danio rerio (Hamilton), is one of the most important vertebrate model organism in genetics, developmental biology, neurophysiology and biomedicine (Rubinstein, 2003; Amsterdam and Hopkins, 2006). It is a small, robust fish, so large numbers can be kept easily and cheaply in the laboratory, where it breeds all year round. Development is rapid, with precursors to all major organs devel...

Pharmacological analysis of zebrafish (Danio rerio) scototaxis

The scototaxis test has been introduced recently to assess anxiety-like phenotypes in fish, including zebrafish. Parametric analyses suggest that scototaxis represents an approach-avoidance conflict, which hints at anxiety. In this model, white avoidance represents anxiety-like behavior, while the number of shuttling events represents activity. Acute or chronic fluoxetine, buspirone, benzodiaze...

Functional Characterization of Zebrafish (Danio rerio) Bcl10

The complexes formed by BCL10, MALT1 and specific members of the family of CARMA proteins (CBM complex), have recently focused much attention because they represent a central hub regulating activation of the transcription factor NF-κB following various cellular stimulations. In this manuscript, we report the functional characterization of a Danio rerio 241 amino acids polypeptide ortholog of th...
